Background


The main purpose of the R-DF21 and Subclades research project is the expansion of the DF21 and Subclades Phylogeny by the discovery of new SNPs and the study of their possible points of origin.

Five of the SNPs that are the focus of this project were found via analysis of whole genome sequences available from the
1000 Genomes Project, with additional SNPs from FTDNA Walk Through the Y (WTY) tests. They are located under the major clade R-L21 and the precise structure can be seen in the haplotree at the bottom of the page.


Note that DF21 is a subclade under L21--two different SNPs that have the unfortunate coincidence of having of the same numerical suffix.

WTY Test Results
  • Kit 20437, DF5+, found three new SNPs, L625, L626, and L627, which are located under R-DF5. L627 was derived for both Reynolds and Cooper (TMRCA ~800 years ago) and ancestral for other families so far, so it is unknown how widespread it might be. L625 and L626 are private to the Reynolds family, and there is no reason for anyone else to order them.
  • Kit 120655, DF5+,  found one new SNP, L658, which is located under R-DF5. L658 may be private.
  • Kit 26411,  DF21+ Z246-, found two new SNPs, L641 and L642, which are located under R-DF21. L641 and L642 may be private.
  • Kit 53766, DF21+ Z246-, found one new SNP, L720, which is located under R-DF21. L720 is not private.
The Irish genome described in
Sequencing and analysis of an Irish human genome is DF21+ DF25- DF5-.

Two samples from the 1000 Genomes project are DF5+.

TMRCA estimates by MikeW using Ken Nordtvedt's Generations5 spreadsheet suggest that R-DF21 is comparatively quite old, with a TMRCA of 3112 YBP versus 4135 YBP for all of R-L21.


DF21 and all the SNPs underneath it can be ordered à la carte from FTDNA; select "Order Advanced Tests" after clicking on "Order Tests and Upgrades" in the left-sidebar. Do not select "Order Advanced SNP Test." Note that only P314.2 is on the "official" FTDNA haplotree and included in the Haplogroup R Deep Clade.

Project members are strongly encouraged to upgrade to 111 markers if they can; there are STR clusters in this project that are only discernible if Y111 has been purchased. 


Anyone who is DF21+ is also encouraged to join the R-L21 project.  If you are L21+ and D21-, you are strongly encouraged to join the R-L21 project -- that is the easiest way for us to track DF21- results.




What Should You Order?

If you have questions about joining the project, or which SNP(s) to order, please contact the project administrator for assistance.


The Deep Clade test does not include any of the new SNPs and generally is not what one should order.


If you are L627+, you may choose to order:

  • L626  Private SNP. Likely candidates for L626+ will have a 62/67 (or better) match with either kit 20437 or kit 66345.

  • L625  Private SNP. Likely candidates for L625+ will be L626+ and be a descendant of Charles W Reynolds, b 1842, Tennessee.
If you are DF5+, you may choose to order:
  • L627  Likely candidates for L627+ will have a GD=24/111 or less with the Cooper-Reynolds cluster, 21-1216.

  • L658  Likely candidates for L658+ will have a GD=24/111 or less with the Cain cluster, 21-1169-A. May be private.
  • Z248  The position of Z248 is uncertain wrt DF5, see below.
If you are DF25+, you may choose to order:
  • DF5  The majority of DF25+ results will also be DF5+.

  • Z248  The position of Z248 is uncertain wrt DF5, see below.
If you are Z246+, you may choose to order:
  • DF25  It is unknown at this time what proportion of DF21+ will also be DF25+. All known DF25+ results have DYS643=9 and match a minimum of two values for DYS717=20, DYS549=11, and DYS504=16. If all three do not match, the third value will be off by a distance of no more than one.

If you are P314.2+, you may choose to order:
  • L362  All results to date that are P314.2+ L362+ belong to the 21-1113-P12-A variety.

If you are DF21+, you may choose to order:
  • P314.2  P314.2+ has distinctive off-modal signatures, please check the deep ancestral variety information below before ordering.

  • Z246  It is unknown at this time what proportion of DF21+ will also be Z246+.
  • L720  It is unknown at this time what proportion of DF21+ will also be L720+.
  • L641  May be private.
  • L642  May be private.
The exact position of Z248 is not known.; it is located at approximately R-DF5. Please order if you are interested in helping to determine the exact position of Z248. This SNP should be regarded as "under investigation."

If you have not tested any of the above SNPs, start by ordering DF21. Please check the haplotree and cluster lists below to see if you fall into a clade or cluster that has a known DF21 status. If you are not in a known clade or cluster, given the age of DF21 and genetic distances among known DF21+ samples, it is not possible to reliably predict who might be derived for DF21; roughly 26% of the DF21 tests to date have been positive. R-DF21 does not have a distinctive STR signature, the R-DF21 modal values are the same as the R-L21 modal values.
